Description: The human MAGE genes are expressed in a wide variety of tumors but not in normal cells, with the exception of the male germ cells, placenta, and, possibly, cells of the developing embryo. Aberrant MAGE expression lead to tumor-specific antigens recognized on a human melanoma by autologous cytolytic T lymphocytes. The human MAGE genes are located on the X chromosome, in four clusters denoted MAGE-A, B, C, and D mapping at q28, p21.3, q26, and p11.2, respectively. Although MAGE-D1 shares strong homology with other members of the MAGE family, but in contrary to these, MAGE-D1 is expressed in almost all normal adult tissues and has been demonstrated to interact with the p75 neurotrophin receptor and to facilitate nerve growth factor-dependent apoptosis. MAGE-D1 also interacts with members of the Dlx/Msx homeodomain family and to regulate the transcriptional function of Dlx5.
