Description:
The human MAGE genes are expressed in a wide variety of tumors but not in normal cells, with the exception of the male germ cells, placenta, and, possibly, cells of the developing embryo. Genes of the MAGE family direct the expression of tumor-specific antigens recognized on a human melanoma by autologous cytolytic T lymphocytes. The MAGE genes are located on the X chromosome, in four clusters denoted MAGE-A, B, C, and D mapping at q28, p21.3, q26, and p11.2, respectively. Twelve closely related MAGE-A genes including MAGE-A2 are located in the Xq28 region. They share 60-98% nucleotide identity in their coding region. The function of these genes remains unknown. The MAGE-A2 gene is expressed in a higher proportion of melanoma tumors than MAGE-A1. It is also expressed in many small-cell lung carcinomas and other lung tumors, laryngeal tumors, and sarcomas. No MAGE-A1 and MAGE-A2 gene expression was found in a large panel of healthy adult tissues, with the exception of testis.
